How Can I Re-assign A Multimedia Keyboard Calculator Button To A Different Calculator App.

Sure thing, here's how:

  1. Right-click desktop and choose New > Shortcut
  2. Type calculator: in the box (including the colon) and then Next
  3. Name your shortcut Calculator (or whatever you want) and Finish
  4. Right-click the new icon and go to properties to set your keystroke (I use Ctrl+Alt+C to open calculator)

This works only first time. After restarting my computer it doesn't works.

Further, yesterday Windows 10 insider preview build 14367 was announced in which following lines were written.

"New keyboard shortcut for Feedback Hub on PC:

We love getting your feedback, and want to make it even easier for you to get into Feedback Hub on PC builds. To capture a screenshot and launch Feedback Hub please use they keyboard shortcut Windows key + F. This opens Feedback Hub and attaches a screenshot of your PC (you can review and remove this before you submit). The old PC keyboard shortcut of Windows key + SHIFT + ? will stop working because of this change."

Therefore this method is not going to work after the anniversary update is arrived.

It there any other method to do this.

The Anniversary update is not removing keyboard shortcuts from desktop shortcut LNK files.  Also, it should not stop working after reboot.  Can you please tell me if the shorcut's properties still reflect the key combination you used, or is it empty now?

Shortcut's properties reflect the key combination but I haven't put the shortcut on desktop. It is in my document folder because I don't keep any icon on my desktop, it is totally empty.

This only works if you put it on the desktop but if you don't want to see it just Mark the icon is hidden.
